Abstract

Research Supplement to Promote Diversity for Toolkit for Fast Multipurpose and Inducible Bioorthogonal Chemistry The goal of this supplement to R01 GM132460 will be to extend the toolkit being developed under the parent award to intersect with the goals of PA-23-189 by funding the research of graduate student Ashley Sias. As an extension of the parent R01 Aim 3, the goal of this proposal is to develop a diverse set of new chemical tools centered on Tetrazine Ligation—the fastest bioorthogonal reaction. The technology developed in this proposal will improve the practicality, generality, tunability and scope of tetrazine ligation. We propose to develop efficient catalytic methods for ‘turning on’ and applying rapid bioorthogonal chemistry in biological context. A mentoring plan and timeline are described that includes various mechanisms for research and career development that will be supported by a mentoring team with both industrial and academic experience.
