Proteomics Core

ABSTRACT, Proteomics Core (PC) The Indiana University Simon Comprehensive Cancer Center (IUSCCC) Proteomics Core (PC) provides state- of-the-art mass spectrometry-based quantitative and qualitative proteomics analyses. We have established standardized methods for sample preparation and mass spectrometry analysis for global untargeted proteomics, phosphoproteomics, thermal proteome profiling, and targeted mass spectrometry analysis of specific targets of interest. In the current grant period, we have established the infrastructure and technical expertise to perform analyses of >100 samples for a single project by discovery proteomics. This work will be extended to targeted proteomics studies in the next funding period. With IUSCCC support, we are also implementing the Olink Explore technology as a joint service between the PC and the Genomics Core. Olink has focused its targeted proteomics target selection for its Explore platform to proteins that are present in human biofluids such as whole blood in a wide dynamic range. Olink Explore has become a highly complementary approach to mass-spectrometry-based discovery proteomics; by offering both types of proteomics services, we will be able to meet the needs of all IUSCCC members over the next grant period. Additional efforts include ongoing and new development of targeted proteomics assays for proteins of interest for IUSCCC users. We will also continue our efforts to provide education and training for IUSCCC users and members of their laboratories on all established and emerging technologies provided by our Shared Resource.
