Abstract

ABSTRACT This supplement grant seeks to address critical equipment limitations hindering our research on receptor-mediated glucose sensing in yeast, which is essential for understanding cellular energy balance and its implications for metabolic diseases like diabetes and obesity. To advance our study, we request funding for four key pieces of equipment: a Fluorescent Microscope to enable real-time visualization of glucose receptor dynamics in live yeast cells, allowing us to track receptor behavior in response to varying glucose levels; a Multimode Microplate Reader to facilitate high-throughput quantification of glucose uptake using the fluorescent glucose analogue 2-NDBG, enabling efficient analysis across various experimental conditions; Protein Extraction Equipment, including a cell homogenizer, ultracentrifuge, chromatography refrigerator, and water purification system, to enhance our ability to extract glucose receptors and transporters from yeast cell membranes for detailed biochemical analysis; and a qRT-PCR System to improve gene expression analysis, allowing us to identify new signaling pathways regulated by glucose receptors Rgt2 and Snf3. The acquisition of these tools will eliminate critical research bottlenecks and significantly enhance our ability to investigate glucose sensing mechanisms, driving the progress of our study and offering insights into the molecular basis of metabolic diseases. 1
